Senolytics: The Drugs That Clear Zombie Cells From Your Body

01 The Zombie Cell Problem
As cells age, some enter a state called senescence — they stop dividing but resist apoptosis (programmed cell death). These 'zombie cells' accumulate with age, secreting inflammatory molecules (the SASP — senescence-associated secretory phenotype) that damage surrounding healthy tissue. By age 60, senescent cells may comprise 10-15% of certain tissues. Removing them has become the most active area in longevity drug development.
02 The First Senolytics
The first senolytic combination — dasatinib (a leukemia drug) + quercetin (a plant flavonoid) — was identified in 2015. It selectively kills senescent cells while sparing healthy ones. In mice, it restored physical function, extended lifespan, and reduced multiple age-related diseases simultaneously. Human trials have shown promising results in diabetic kidney disease, idiopathic pulmonary fibrosis, and osteoarthritis. 12 clinical trials are now ongoing.
03 The Challenge
Current senolytics are 'hit and run' — they're taken intermittently (e.g., 2 days per month) to clear senescent cells without causing excessive toxicity. The challenge is that senescent cells aren't all bad — they play roles in wound healing, tissue regeneration, and tumor suppression. Removing too many, or removing them from the wrong tissues, could be harmful. The next generation of senolytics aims for tissue-specific targeting: clearing senescent cells from fat tissue (to improve metabolism) while leaving skin senescent cells intact (for wound healing).
